I wonder if the frequency of jacking it from the rear diff or leaving it jacked from there makes a difference?

Most folks would only do that a couple of times a year I would think. Rotating tires, maybe rear brakes, etc.

Once you put stands under it, then drop the jack to take the pressure off the diff, I shouldn't think it would hurt.
